Production of recombinant proteins in plants

In this BioRN project (http://www.biorn.org/), a plant expression platform will be developed in which the defense mechanism against transgene over-expression (post-transcriptional gene silencing or RNA interference (RNAi)) will be inactivated. This will be achieved by down-regulating the activity of the RNA-directed RNA polymerase 6 (RDR6). In an RNAi-deficient background, significant transgene transcription levels are expected. However, to further increase transgene expression the efficiency of translation will be enhanced by introducing a viral-derived 'translational enhancer sequence' (TES) at the transgene 5' ends. Using inducible and/or tissue specific promoters, the production and the targeting of recombinant proteins can be further optimized.

Tobacco plants produce a high biomass and are therefore suitable as "bioreactors".
