Objective is the development and breeding of resistant rootstocks using classical plant breeding. In this project, apple proliferation -resistant wild types (Malus sieboldii and Malus sieboldii derived hybrids) are crossed with agronomically interesting rootstock cultivars. Investigations at AlPlanta-IPR include the development of molecular markers for marker -assisted selection, the genetical analysis of natural resistance as well as resistance screening in vitro and in vivo. The in vitro grafting method, which has been recently established, is a highly efficient inoculation method which offers a rapid resistance screening. Using quantitative real- time PCR, a fast evaluation of the resistance of inoculated genotypes is possible. Real- time PCR is also adopted to study the differential gene expression in resistant versus susceptible genotypes or in infected plants versus healthy plants of the same genotype.
